چکیده
Determination of the reinitiation time We define a single translocation event as the time from when the DNA end-to-end distance starts to decrease due to translocation activity until the point where the end-toend distance returns to its resting length due to termination of translocation. Considering only one of the translocation directions of a bidirectionally translocating enzyme, the reinitiation time Treini,1 = Tbind,HsdR + Tini is simply the time between the end of one translocation event and the start of new translocation event in that direction. The average measurement time to observe N1 translocation events 〈Tmeas〉 is then ( ) event,1 reini,1 N1

Dynamics of initiation, termination and reinitiation of DNA translocation by the motor protein EcoR124I.
Type I restriction enzymes use two motors to translocate DNA before carrying out DNA cleavage. The motor function is accomplished by amino-acid motifs typical for superfamily 2 helicases, although DNA unwinding is not observed. Using a combination of extensive single-molecule magnetic tweezers and stopped-flow bulk measurements, we fully characterized the (re)initiation of DNA translocation by ...
متن کاملTranscriptional Coactivator CBP Facilitates Transcription Initiation and Reinitiation of HTLV-I and Cyclin D2 Promoter
HTLV-I is the etiologic agent for adult T-cell leukemia/lymphoma (ATL) and HTLV-I-associated myelopathy/tropical spastic paraparesis (HAM/TSP). Taxi, the major activator of this virus, is a 40- kDa (353 amino acid) phosphoprotein, predominantly localized in the nucleus of the host cell, which functions to trans-activate both viral and cellular promoters. Recently it has been shown that HTLV-I a...
متن کاملStability of RNA chain elongation complexes formed with RNA polymerase and denatured DNA templates.
A random transcription system was used to analyze RNA chain-elongation complexes with RNA polymerases I and II from mouse ascites sarcoma cells. The molecular size of synthesized RNA was analyzed to characterize the elongation reaction by RNA polymerases. The RNA chain-elongation complex after initiation of RNA synthesis on denatured DNA by RNA polymerase II was not decomposed with heparin whic...
متن کاملTranscription termination factor La is also an initiation factor for RNA polymerase III.
La RNA-binding protein is a transcription termination factor that facilitates recycling of template and RNA polymerase (pol) 111. Transcription complexes preassembled on immobilized templates were depleted of pol III after a single round of RNA synthesis in the presence of heparin and sarkosyl. The isolated complexes could then be complemented with highly purified pol III and/or recombinant La ...
متن کاملA subcomplex of RNA polymerase III subunits involved in transcription termination and reinitiation.
While initiation of transcription by RNA polymerase III (Pol III) has been thoroughly investigated, molecular mechanisms driving transcription termination remain poorly understood. Here we describe how the characterization of the in vitro transcriptional properties of a Pol III variant (Pol IIIdelta), lacking the C11, C37, and C53 subunits, revealed crucial information about the mechanisms of P...
